What would be instrumental in identifying gemstones in jewelry? Is it necessary to take the gemstone out of the setting to identify ( besides diamonds)? Thank you.

It is not necessary to take gems out of mountings to identify them.....even diamonds, unless you wish to send the diamond to the GIA.

With standard gemological tools, you can check a mounted stone:
Polariscope, singly doubly refractive possible optic figure.
Refractometer: If the table of the gem is above the prongs, one can get a refractive index.
Spectroscope: Shouldn't be a problem with mounted goods.
SWLW UV: no problem
Microscopic Observation: Again, no problem

And if you are fortunate enough to own a GemmoRaman, well, enough said. :D

Like Barbra sayd, can be done in setting. You only take out a gem from setting if you want to know precise carat weight of said gem.
